Objective1 To investigate the plasma levels of gonadotropins and gonadal hormones secretion in man with prostate cancer.2 To investigate the effects of overexpression of LRP16 on proliferation of human prostate cancer ALVA41 and DU145 cells .3 To investigate the possible mechanisms of the effects of overexpression of LRP16 on proliferation of human prostate cancer ALVA41 and DU145 cells .Methods1 Fifty-three men with prostate cancer and 253 normal controls were recruited for this study. The levels of gonadotropins (LH and FSH ) and gonadal hormones (Tand E2) before treatment were determined.2(1) LRP16 eucaryotic expression vector (pcDNA3.1-LRP16) was constructed and transfected into ALVA41 and DU145 cells with the control pcDNA3.1(+) vector, respectively, and then four groups of cells expressed LRP16 gene or empty vector (A16, A3.1, D16, D3.1) were selected by G418 and cultured for further study;(2) Growth curve of each group was measured with MTT method.3 Total RNAs extracted from two kinds of cells were determined for estrogen receptor a (ERa), p (ERP) and androgen receptor (AR) expression by RT-PCR.Results1 T, TSI(T/LH) and FSH levels in the prostate cancer group were low compared with the control group (P<0.05). But there was no significant difference of E2 and LH levels between prostate cancer and control groups(P>0.05).2 The diagnosis standard of T is 15nmol/L.3 The diagnosis standard of TSI is 4.0.4 (1) Overexpression of LRP16 stimulated DU145 cell proliferation, the ratios of folds between D16 and D3.1 cells in 24, 48, 72, 96 and 120 h were 1.034(P>0.05 ) , 1.100 (p<0.05), 1.127 (p<0.01), 1.626 (p<0.01) and 1.548 (p<0.01), respectively;(2)Overexpression of LRP16 had no influence on the proliferation of ALVA41 cells , the ratios of folds between A16 and A3.1 cells in 24, 48, 72, 96 and 120 h were 0.998,1.040,0.944,1.018 and 1.036 (all P values >0.05).5 ERa and ERp were expressed in both ALVA41 and DU145 cells, AR was only expressed in ALVA41 but not in DU145 cells.Conclusion1 Prostate cancer patients have low levels of total and free testosterone.2 The incidence of prostate cancer may be elevated when T or TSI is less than the diagnosis standard.3 Overexprssion of LRP16 Stimulates prostate cancer DU145 cell proliferation.4 Overexprssion of LRP16 Stimulates prostate cancer DU145 cell proliferation, and this effect may be not directly mediated by AR.
