Objective: Investigate the expression of Discoidin domain receptors (DDRs) in keloids. Method: The expression of DDR and its downstream signaling pathways were detected by IHC, WB, RT-PCR and FQ-PCR. The effect of DDR on proliferation and apoptosis of KFb were determined by the MTS assay and FCM technique. The potential of DDR to stimulate collagen synthesis in the KFb were determined using the method of [~3H] PI. Effects on transferring DDR-ASODN to KFb were also studied. Results: The expression of DDR was showed significantly higher in KFb than NFb using IHC, FQ-PCR, WB and RT-PCR. DDR's downstream signal molecules were also highly expressed. The result of the MTS assay shows that DDR enhance the proliferation of KFb. Applying the DDR-ASODN to KFb shows that higher apoptosis and less expression of P53, MMP-1 and MMP-2, the collagen synthesis were also descended. Conclusions: The expression of DDR mRNA and protein in KFb were at high level compared to NFb. DDR enhance the proliferation of KFb and increase the collagen protein secretion. Transfection of DDR-ASODN into KFb were increased the apoptosis of KFb, descended the collagen synthesis and the expression of MMP-1, MMP-2 and P53. These findings suggest that DDR may play an important role in the pathogenesis of keloid.
