Clinic And Gene Expression Study On Radiosensitization Of Fuzheng Zengxiao Formula In NSCLC

【Objective】Fuzheng Zengxiao Formula is an experiential formula created by Prof. Zhang Daizhao. Former clinic and experimental studies testified its radiosensitization. Fuzheng Zengxiao Formula , combined with radiotherapy , whose influence on short-term effect and quality of life in non-small cell lung cancer (NSCLC) were observed in the paper's prospective clinic study which was based on former studies. Experimental part was to investigate its radiosensitizing mechanism and effective target on genic level by gene chip.【Methods】It was a prospective, random and parallel clinic study design.62 cases of NSCLC were divided into observe group (Fuzheng Zengxiao Formula combined with radiotherapy), 32 cases and control group (radiotherapy only), 30 cases. Short-term curative effects, side effects and quality of life were observed.PAa cell hairless mice models were erected in experimental study and divided into four groups:Ⅰ(blank group),Ⅱ(radiotherapy group),Ⅲ(Chinese medicine group, Formula and radiotherapy),Ⅳ(Western medicine group, metronidazole and radiotherapy). radiotherapy ofβelectron ray (groupⅠhad no radiotherapy) was given when tumors'阿average diameter was about 1 centimetre.23 hours later, got the tumors quickly, abstracted and separated mRNA, then finished reverse transcription and biotin labeling. Affymetrix Human Genome U133 2.0 Array Gene Chips were adopted. Real-time PCR was to validate the reliability of scanner results.【Results】Clinic study showed short-term effective rates of two groups was 68.75 percent and 39 percent, the rate of observe group was higher than that of control group, P<0.05.The level white blood cell, lung and skin symptoms in observe group were better than those of control group, P<0.05. KPS of observe group was higher than that of control group, P<0.001. Observe group was better than control group on whole health, weakness, difficult breathing and cough in EORTC QLQ-C30 and QLQ-LC13.857 genes were differently expressed between the group received the Formula besides radiotherapy and the group only received radiotherapy, including 349 up-regulated genes and 508 down-regulated genes.
