Allergenic nasatitis (AR) is allergenically inflammatory disease of type I induced by allergen, which is by The meThod of IgE and is characteristic by The esophocyte, mast cells, T lymphocyte. The symptom is nasal itch, sneeze and snivel wiTh The complication of asThma. The abroad study revealed The occur of allergenic nasatitis is closely related to economic development. Recent years, The mobidity have increased significantly to 10% wiTh serious damage to The healTh of people.AR is treated by modern medicine wiTh immunal Therapy or drug, which have a lot of shortcoming. For immunal Therapy, The allergenic antigen is difficult to be determined, The effect is slow to occur, The course is too long to finish for The patients, The disease is difficult cured. The drug treating Allergenic nasatitis consist of drug anti-histamine, cortisol, atropine, wiTh rapid effect and more side effect.AR is called "biqiu", "qiuti" in Traditional Chinese Medicine. External Application of Herbal Drug on Acupoints (EAHDA) treat The AR wiTh long time and good results. But The immunal mechanism is undermined. In our study, The immunal mechanism of EAHDA treating The allergenic nasatitis in investigated, which provide scientific information for Therapy.1 .MeThod and content1.1 published articles are studiedThe published articles are studied to declare The clinical and experimental investigation and provide The prospection.1.2 experimental studyIn our study, The effects of EAHDA on The AR are investigated in motivation, paThology, mast cells in blood or BALF, eosinophilia, IgE, lymphocyte specific proliferation, specific lymphocytokine secretion.1.2.1 60 BALBA/c murine wiTh 7 weeks old, SPF and 20-25g weight are randomized given into The following groups: The blank of normal murine, The experimental group of AR murine treated by EAHDA, The positive group of AR murine treated by cortisol, The negative group of AR murine treated by PBS, The group of AR murine.1.2.2 The murine wiTh AR are prepared by intraperitoneally injection of OVA.1.2.3 The EAHDA are prepared according to The Traditional Chinese Medicine Therapy.1.2.4 For The murine before and after The treatment are observed, The immune response of blood or respiratory passage including The cells classification, IgE, Thl/Th2 lymphocyte and The secreting lymphocyte, The mast cells and The degranulation, are investigated and analyzing.2.The results2.1 AR are treated by modern medicine have many side effect, which is The most serious problem. The oTher treatments wiTh less shortcoming and significant effect are urgent. EAHDA provide a good way for AR. In anoTher aspect, modern medicine offered good meThod and Theory for EAHDA. EAHDA also have some shortcoming, including The complicated and long period of preparation. The effect was not steady, sometime its effect was prominent, some its effect was not very good. In more word, some patients can not insist to receive EAHDA. More reports on EAHDA treating AR are published and The effects are very prominent. However, The oTher reports were repeated. In fact, There were many meThod on AR treated by EAHDA pffered a good inspiration. By new achievement in modern molecular biology, molecular immunology, we can systematically study AR on cell level and offer scientific and objective foundation to AR treatment.2.2 Compared wiTh The normal murine in The blank group, There were more number of sneeze for The AR murine group;There were less number of sneeze for The AR murine treated by EAHDA compared wiTh Those AR murine. Our study showed That AR murine was successfully modeled, murine obvious appeal: increasing number of sneeze, The average of sneezes in group of treatment by EAHDA was significant less Than That of group of AR murine motivated by OVA wiThout treatment.2.3 According to our study The changes of paThology on nose mucosa and aspiratory airway in models murine. The result was That nosal mucosa in normal groups had some mild eosinophilia. However, The oTher cells in murine of modeled of OVA group, were moderate degree, especially eosinophilia was more Than That of murine of normal group.2.4 To test of intraperitoneal degranulation of mast cells occurred in OVA group, which was prominent difference between in and normal group. Comparing wiTh OVA group, degranulation in application group murine and cortisol group prominently decreased, which had statistical significance.2.5 One of immunal response in murine blood system, cell classification in blood, wediscovered That The Eos absolate level and percent in OVA group increased significantly compared wiTh normal murine, which had statistical difference. Compared wiTh OVA group murine, murine in application group and cortisol group, The Eos absolate level and percent decreased significantly.2.6 Serum IgE study showed That IgE level in murine of OVA group and PBS group obviously increased compared wiTh Those of normal murine;Compared wiTh OVA group murine, murine in application group and cortisol group, The IgE level in murine of OVA group and PBS group obviously increased compared wiTh Those of normal murine.2.7 We discovered That serai Thl lymphocytokine such as IFN- y , IL-2 from murine in OVA group were significant lower Than Those from murine in normal group(P<0.05), Th2 lymphocytokine such as IL-4, IL-4 from murine in OVA group were significant higher Than Those from murine in normal group (P<0.05), The change of IFN- Y, IL-4 were greater significant Than Those of IL-2, IL-10. After treatment of EAHDA or cortisol, The serai Thl lymphocytokine of murine decreased, The serai Th2 lymphocytokine of murine increased.2.8 The number of lymphocyte, macrophage, eosinophele, neuphocyte from The BALF in The OVA group are higher Than Those in The normal group(P<0.05), (PO.05), The change of eosinophele is most significant;The number of above cyto from The BALF decreased after The treatment of The EAHDA or cortisol, The eosinophele is also most significantly decreased.Compared wiTh The change of lymphocytokine in BALF or serum, The secretion of lymphocyte from The spleen lymphocyte are more higher and change more significantly. 3. Conclusions3.1 There were some similarity in experimental and clinical study between AR and Allergic asThma.3.2 AR murine treated by EAHDA or cortisol, The sneezes number decreased significantly.3.3 The eosinophilia in nasal mucosum of The murine from The AR group was very distinct. Eosinophilia plays a important role in paThogenesis for AR.3.4 For The AR murine treated wiTh EAHDA or cortisol, degranulation of The macrophage were significantly decreased Through inhibiting The secretion of histamine, 5-HT and oTher allergic cytokine, decreasing The exudation of inflammatory media. We presumed That EAHDA or cortisol treat The allergic diseaseby The meThod of stabilization of mast cell membrane, inhibition of degranulation of mast cell, decreasing production of inflammatory media.3.5 In allergic reaction of type of I caused by OVA, immunal paThologic infiammating response in respiratory airway, eosinophilia mainly play an important part in AR murine motivated by OVA. EAHDA or cortisol, EAHDA or cortisol worked by restricting The production of eosinophilia in blood.3.6 OVA group and PBS group murine spleen lymphocyte were excitede by OVA in body, when They were excited again out body, OVA specific lymphocyte clone prominently activated, which acted significantly proliferation. AR murine handled wiTh PBS can not inhibited OVA specific proliferation. AR murine treated by EAHDA or cortisol inhibited immunal cyto, made it never activated when were excited by OVA out body, which acted no obvious proliferation.3.7 Immunal system of AR murine activated by OVA showed specific body humoralimmune response-------specific IgE, which indicated That AR activated wiTh OVAwas allergic reaction of type I Thorough of IgE, IgE effected paThogenesis of AR activated wiTh OVA and also indicated That AR treated by EAHDA or cortisol wiTh decreasing specific Ing in murine body.3.8 Thl/Th2 lymphocytokine in AR murine activated wiTh OVA lost balance and by EAHDA or cortisol EAHDA or cortisol can correct it.3.9 There was different level of IFN-Y, IL-2, IL-4 and IL-10 in BALF compared wiTh Those in serum. The primary reflected Thl/Th2 lost balance in respiratory airway, which acted in The paThogenise of AR murine activated wiTh OVA, relative lymphoid tissue in bronchia and local lymphodus did not affect BALF. It is The local immune reaction, which was respitatory airway action of Allergic Rhinitis.
