Objective: Sterigmatocystin (ST) is one of the most common seen contaminating mycotoxins in the foodstuffs of high incidence area of cancers in China. The carcinogenic and immunological effects of ST on experimental animal and human cells have been reported.The damaging effects and mechanisms of mycotoxin on brain have caused the concern of sicentists all of the world, but putative effects and mechanisms of sterigmatocystin on brain has not been seen in the literature. Circumventricular organs(CVOs)are some miniorgans around cerebral ventricles that include subfornical organ, median eminence, organum vasculosum of lamina terminalis, choroid plexus, subcommissural organ, pineal body and area postrema etc. There is no blood-brain barrier in CVOs and thus , they are ideal sites in the monitoring of components of blood. The aim of this study is to explore the putative effects of sterigmatocystin on CVOs by single oral administration of ST at different time point. Ultrastructural changes of ependemal cells of subfornical organ, choroid plexus, ependemal cells of lateral recess of the fourth ventricle and the structure under ependyma were observed with scanning and transmission electron microscopic method. The expression of TNF-αand TGF-β2 at mRNA level in subfornical organ, choroid plexus, lateral recess of the fourth ventricle and other parts of brain were detected by in situ hybridization. The apoptosis in subfornical organ, choroid plexus, lateral recess of the fourth ventricle and other parts of brain were detected by TUNEL. This study was first investigation in effects and mechanisms of sterigmatocystin on CVOs that established invariably the foundation for effects and mechanisms of sterigmatocystin on brain in future.Methods:...
