Objective1 To investigate the state of p16INK4a promoter methylation in psoriatic epidermis .2 To analyze the relationship between p16INK4a promoter methylation and the expression level of p16INK4a mRNA in epidermis from patients with psoriasis.3 To explore preliminarily the relationship between p16INK4a promoter methylation and immunologic abnormality in psoriasis.4 To explore the effect of promoter methylation in mechanisms of regulating p16INK4a expression in psoriasis.Methods1 The DNA and RNA samples of epidermis were obtained from 56 plaque psoriatic lesions, 28 non-lesions, and 38 healthy persons as the control group.2 The severity of disease was evaluated by psoriasis area and severity index (PASI).3 The p16INK4a promoter methylation was detected by methylation specific PCR (MSP),and the correlation between methylation and clinical conditions was assessed in psoriasis.4 Reverse transcriptase polymerase chain reaction(RT-PCR) technique was applied to detect the expression level of p16INK4a mRNA,which in order to investigating the relationship between p16INK4a promoter methylation and the expression alteration of p16INK4a mRNA in epidermis.5 The expression levels of IFN-gamma receptor mRNA and TNF receptor mRNA were detected by RT-PCR technique , to analyze the relationship between p16INK4a promoter methylation and the expression level of two receptors in the psoriatic epidermis.6 The deletions and mutations of exons (1α ,1β and 2) of CDKN2A gene locus were detected by polymerase chain reaction (PCR)-single strand conformation...
