Application of self fat tissue has been widely accepted in treatment for soft tissue defect. However, either pedicled or free adipose tissue has its disadvantages. On the one hand, the transferring of autologous fat for massive defects takes the price of operative injury and donor site deformity. On the other hand, repeated injections are required for minor defects because of a high absorbation of injected fat. Tissue engineering, as a new emergencing and promising technology focused on biomedical and biomarterial science, is predominant in forming autotissue mass by cultivating and proliferating small amount of seed cells in vitro. It is not until recently that adipose tissue engineering has aroused great interest in restoring soft tissue defects. The integration ofthe three major ingredients ---- seed cells, microcarriers andmicroenviroment, will induce the formation of new tissue which will be used in the treatment for tissue or organ defect.Objectives1. To isolate and cultivate preadipocytes from human adipose tissue.2. To screen suitable scaffolds as well as to investigate the proper microenviroment for adipose tissue engineering. To investigatethe feasibility of two commonly used scaffolds----the calcium alginateand collagen sponge for human preadipocytes.
