| Objective: To study the effects of ElA gene on the growth rate, tumorigenicity and radio-chemosensitivity of human lymph node metastasis cell line LN686 derived from the patient with human head and neck squamous cell carcinoma(HNSCC), and make a preliminary study for gene therapy of lymph node metastasis of HNSCC. Methods: The recombinant -pcDNA3. 1-E1A plasmid containing the 1. 77Kb ElA gene, was introduced into the metastasis cell line —LN686 by using liposome. The cells resistant to G418 were selected and LN686-vect (containing empty vector) and LN686-E1A (transfected with pcDNA3. 1-E1A) cells were obtained. The changes of growth rates, doubling times, and cell cycle were also studied. After LN686 LN686-vect and LN686-E1A cells were injected into nude mouse, the time of tumor formation, growth rates and weight of tumor were observed. LN686, LN686-vect and LN686-E1A cells were treated with cisplatin, paclitaxel , bleomycin , 5-FU for 24h and irradiation, respectively. Viable cells were quantitated by MTT assays.
