One approch to investigating human carcino-genesis iε to transform normal cells into malignant ceils in ritro. However, there are a few reports describing carcinogen-induced transformation of human epithelial cells. One problem in the in vitro transformation of human epithelial cells is the lack of a suitable model.We hare developed a new model which can be used for the investigation of in vitro transformation of human tracheobronchial epithelial cells. The advantages of both organ culture and cell culture were combined in this new transformation assay. Human tracheobronchial tissues were cultured in CMRLl066 medium containing insulin, hydrocortisone. trace elements, with or without fetal bovine serum(FBS) in a rocked atmosphlere chamber and exposed to chemical carcinogens during organ culture.
