Oncogenes (C-erbB1, C-erb B2) amplification and expression in urinary bladder cancer and its clinical significance1.Exfoliated cell C-erb B1, C-erbB2 oncogenes were amplified in 58 patients with urinary bladder cancer by differential PCR(Polymerase Chain Reaction). Cytology of these cells were graded at the same time for early precise diagnosis. The results showed that neither C-erbB1 nor C-erb B2 oncogene was found in Grade G0 and G1 exfoliated cancer cells. In 5 cases of grade G2 in cytology, only 1 case had C-erb Bl amplification and 2 cases had C-erbB2 amplification. In 7 cases of grade G3, 2 had C-erbBl and 5 had C-erb B2 amplification. Differential PCR is a sensitive and quick method to detect C-erb B1 and C-erb B2 oncogene amplification in urinary bladder washing. It provides evidence for more precise gradation of bladder cancer when combined with cytology gradation and this facilitates clinicians to chose appropriate treatment strategy.2. Expression of EGFR and P185 protein was examined in 112 urinary bladder cancer patients by microwave oven antigen retrieval immunohistochemical method(S-Pmethod). The relationship of EGFR and P185 protein expression with the stage, grade, recurrence and prognosis was explored . Our results showed that no expression of EGFR and P185 protein could be detected in 10 normal control bladder tissue.EGFR and P185 protein was expressed in 55.30% and 46.43% bladder cancer tissue respectively.Both were expressed in 40.18% bladder cancer tissue. The expression rate of EGFR and P185 protein in G1, G2 and G3 bladder cancer tissue was 25.00% vs 18.75%, 56.00% vs 44.00% and 86.67% vs 80.00% respectively. EGFR and P185 protein expression in Tis, T1; T2-T4 bladder cancer tissue was 41.67% vs 23.33% and 71.85% vs 73.08% respectively.
