Using Flow Cytometry (FCM), Image Cytometry (ICM) and c-myc gene Polymerase Chain Reaction (PCR) an retrospective study was performed on 41 paraffin-embedded samples of esophageal squamous cell carcinoma. The DNA content of esophageal cancer was measured by FCM and ICM. The results showed that 11 cases (26.8%) were diploidy and the other 30 cases were aneuploidy. A close correlation was observed among the DNA ploidy status, lymph node metastasis, TNM staging and degree of the cancer cell differentiation. DNA ploidy status was also found related to the prognosis of patients (P<0.01) The five year survival rate of diploidy tumors was significantly higher than that of aneuploidy tumors. The percentage of Sphase cells (S%) determined by FCM was related to the prognosis of patients. The survival rate of patients with S%<15% was significantly higer than that with S%>15 (P<0.01). The relative DNA content determined by ICM was revealed in three types of histogram. There were 8 cases (19.5%) in type 1.12 cases (29.3%) in type II and 21 cases (51.2%) in type III. Type I was diplody tumors (19.5%, 8/41), Type II and III were aneuploidy tumors (80.5%, 33/41) The three types of histograms showed certain correlation with TNM staging and degree of tissue differentiation. None of type I tumors (8 cases) showed lymph node metastasis. An obvious correlation was noticed between different type of histogram and prognosis (P<0.01). The best prognosis was observed in type I, then type II and the worst in type III 5C% of aneuploidy cancer was also related to the prognosis of tumors with 5C%<10% was significantly better than that with 5C%>10%. The mean value of ICM determined relative DNA content. Its range was from 1.2 to 7.67. The lower mean value showed a better prognosis and higher for a poor prognosis. A similar pattern...
