| Hypertension is the most prevalent cardiovascular disease with high morbidity and mortality in the world. Blood pressure is regulated by a variety of the mechanisms in which the interactions take place between the products of several genetic loci and a number of environment factors. Little is known the number of genes actually effect on blood pressure, the mode of their transmission, or their interaction with other genes and environment factors. A large of studies has been done trying to figure out the heritable elements related to blood pressure. And it has been detected that more than 30 candidate genes may contribute to the abnormal blood pressure.Because of the well documented role of this system in the control of blood pressure and the pathogenesis of several forms of hypertension, genes of the renin-angiotensin system have been most extensively studied. In this system, renin acts on a single substrate, angiotensinogen (AGT), to form angiotensin I. The latter is further converted Angiotensinogen and angiotensin I-converting enzyme are very important components in the renin-angioten system. The two genes were studied as hypertension candidate genes in our research.-converting enzyme into angiotensin II, one of the most active vascular factors.Angiotensinogen and angiotensin I -converting enzyme are very important component in the reniin-angiotensin system and the gene codon for them are hypertension candidate genes studied in our this research work. The insertion/deletion polymorphism of ACE gene and the polymorphism of M235T of AGT gene are most extensively studied. A number of linkage and association studies have since examined several clinical situation and various population.We first examined the linkage of these two polymorphisms with essential hypertension in Tibetan families. The 42 subjects from 10 Tibetan families were genotyped for ACE gene I/D polymorphism using PCR (polymerase chain reaction) technique and for AGT gene M235T polymorphism using RFLP (restriction fragment length polymorphism) technique. The genetic linkage analysis showed that the LOD value of these two loci have not reached 3.
