Immunoregulators are divided into intensifiers and debilitaters according to their effects on immunity system. The former have the ability to facilitate immunoregulations; the latter, in contrast, to debilitate. GPIF in this study are extracted from goats'placentas with the help of advanced biotechnology, and found to be an immuno-intensifiers. In this article, following the research principles into immuno- intensifiers, we focus on the systematical study of the GPIF's facilitating immunoregulations and their primary mechanism. The study intends to be a pioneer one and exploit the active components of the goat plecenta extract; for the purpose of supplying pre-clinic research for the confirmation of GPIF and experimental resources for GPIF's clinical application and laying foundation for the purification of the functional components and further study of GPIF.The thesis falls into four parts, namely, systematical study repectively on GPIF's effect on T cellular immunity, humoral immunity and macrophages, and partial study on the mechanism of GPIF's immuno-intensifying. On the basis of these studies, tentative study has also been done on the immuno-function of T cellular with GPP-Ⅰ, one of the purified components of GPIF.In the first part, GPIF's promoting T lymphocytes function and its effect mechanism were studied and discussed in vivo and in vitro with research models: (1) the experiments in vitro were based on the normal T lymphocytes,the suppressed T lymphocytes which were caused by cyclophosphamide and 60Coγ-ray irradiation respectively. The MTT method was used to detect the T lymphopoiesis induced by ConA; FACS with immunofluorescence marked with antibody was used to analyze the rate of T lymphocytes that expressed CD3,CD4,CD8,CD28 and CD152; ELISA was used to detect content of IL2 and IFN-γ;the activity of IL-4 was mensurated by inderected method.(2) the experiments in vivo such as DTH , FACS and so on were based on immunosuppressed animal models that were cause by cyclophosphamide and 60Coγ- ray irradiation respectively.
