The cancer cells often share the similarity with stem cells in man characteristics: they all have undifferentiated phenotype; Stem cells can immigrate into other tissues, while cancer cells have metastatic capacity. Both of them can transform into each other under some special conditions. In additionally, multiple mutations are necessary for malignant transformation in oncogenesis, stem cells can accumulate more mutations to cause malignant transformation than differentiated functional cells. It is reasonable to propose that the tumor might arise from stem cells. Researchers have isolated leukaemia cells that have markers of HSCs from acute myeloid leukaemia (AML). Only these rare leukaemia cells can transfer AML from human patients to NOD/SCID mice. It reveals that the cancer stem cells might exist in many tumors, which have both capacity of self-renewal by self proliferation and tumorigenicity. Now the cancer stem cells have been found in many solid tumors, for example breast cancer, human train tumor etc. Primary hepatocellular carcinoma is one of the most malignant tumors in our country. Studies have shown that the oval cells, which have characteristics of progenitor in liver, contribute to hepatocarcinogenesis. The heterogeneity of HCC also revealed that not every liver cancer cells have capacity of initiating tumor, and there are great differences of drug resistance between the liver cancer cells. It can be proposed that the cancer cells might exist in the HCC, which have special phenotype and function. Isolating these cancer cells from HCC and identifying their stem like characteristics can help us validate mechanism of carcinogenesis, recurrence and metastasis of HCC, and a new strategy for HCC therapy will also be found.Hoechst/FACS is a new method for enriching stem cells efficiently.
