Objective: 1. To observe morphology and growing status of Bone Marrow-derived Human Mesenchymal Stem Cells(hMSCs) in vitro, make up an optimum method of culture and isolation of hMSCs, research cellular characteristic of hMSCs and established foundation of tissue engineering, the methods and conditions of harvesting, isolating, cultivating and identifying hMSCs were studied. 2. To select a suitable inducing concentration, the effects of different concentrations of 5-azacytidine inducing hMSCs into cardiomyocytes in vitro were compared, and to confirm the construction, morphology and function of cardiomyocytes, hMSCs induced into cardiomyocytes with 5-azacytidine were identified. 3. To confirm whether endothlin-1 could promote hMSCs to differentiate into cardiomyocytes in vitro, the effects of endothlin-1 in differentiation of hMSCs into cardiomyocytes in vitro were tested.Methods and Results:Part one Isolation, culturing and identifying of hMSCsMethods: the samples came from human bone marrow.
