Neural stem cells(NSCs)is an adult stem cells. They have the powerful plasticity and ability of renew. So it can be used in the research of cell therapy and gene therapy. The aim of this experiment was to establish a suitable culture method for NSCs derived from hippocampus of guinea pig and compare the proportion of NSCs in secondary expanded clones from different culture passage.METHODS â‘ With the presence of mitogenic factors EGF and bFGF (20 μg/L), NSCs was isolated and incubated from hippocampus of newborn (24h) guinea pig with seurum-free incubation technique. Differentiation of NSCs into neurons, gliacytes and Schwann cell-like cells was induced by removing EGF and bFGF from the medium and adding 100 ml/L of fetal bovine serum. Neurospheres were collected and labeled with BrdU(10 μg/ml) in the culture medium.
