Lung cancer is one kind of malignant tumors with the highest incidence and mortality. The early-stage diagnosis for lung cancer will be important to patients for improving their life quality and survival period. The indexes in serum for lung cancer used for clinical diagnosis are not specific, also can be detected in other kinds of cancers and some kinds of benign lung diseases. It is urgent to find the novel and specific indexes for lung cancer diagnosis. The proteomics-based techniques will provide the strong support for high throughput screening tumor markers, including two-dimensional gel electrophoresis (2-DE), mass spectrometry and biological informatics.Tumor serologic proteome analysis (SERPA) is integrative method including proteomics and tumor immunology. It combines the advantage of separating the numerous proteins by proteomics-based technique with characteristic of sensitive immunoreactivity between antigens and antibodies, SERPA can be used to screen plentiful tumor-associated antigens which can be identified with MS. Then tumor-associated antigens can be cloned and expressed through genetic engineering, finally, the potential capacities of the expressed proteins as the diagnosis indexes can be evaluated by detecting their antigenicity through immunoreaction with sera from tumor patients.In this study, SERPA was used to establish differential Western blotting maps including lung adenocarcinoma tissues and mixed sera from both lung adenocarcinoma patients and from patients with benign lung diseases.
