Postoperative contracture remain the most frustruting late complications of skin grafts. Existing medical and surgical strategies to prevent it was limited to increasing the thickness of graft, Application of acellular dermal matrix, growth factors, topical splinter or presure garment etcl. Each treatment presents various drawbacks in terms of effectiveness, time, cost, discomfort, and inconvenience, and can't spread widely in clinic.In the 1970s, Gabbiani discovered that the Myofibroblast(MFB) is the engine of wound contraction, After that, Rudoph and Jerry detected that full-thickness skin grafts inhibit wound contraction more successfully than split-thickness skin graft by more effectively suppressing the life cycle of the MFB. In 2002, luan and colleagues proved that papaverine can permeate through skin and reach high effective concentration in skin tissue to inhibit the fuction of MFB in fibrous capsuie. Inspired from luan's finding, we proceeded (?) experimental study on inhibition of skin graft contraction with papaverine cream to search a safe effective simple and reliable method to promote the effect of skin grafts. The experiment obtained good result, but the permeability of papaverine hydrochloride in skin graft is not evaluated and the mechanism of action is not revealed intensively, So we do the further reserch in this study.Firstly, we detected the concentration of papaverine in the experimental group and control group with spectrophotometer to ascertain the active drug concentration. The result indicate that in vivo, the papaverine concentration was 36-5 μg/ml in skin grafts within 12h when 0.25g/cm~2 papaverine cream was externally used. There is no papaverine was detacted in coutrol group.Secondly , The size of the graft area in each group was measured. The result showed that external using of papaverine can inhibit the skin graft contraction effectively. This effect mainly localized in the forty days after operation, and when stop using of papaverine on 90 days after operation, the difference between two group contraction rate is not...
