Pancreatic carcinomas are among the carcinomas with the worst prognosis with 5-year survival rate less than 5%. Despite considerable progress has been made in the understanding of the molecular biology of pancreatic carcinoma, the molecular mechanism of pancreatic carcinogenesis remains to be exploited. Point mutation of the K-ras gene is very common in many human cancers, such as colorectal cancer, lung cancer and pancreatic adenocarcinoma. Pancreatic adenocarcinoma shows the highest frequency of K-ras gene mutations among common human cancers; but the reasons are unknown.Recently Wang et al. found that 18 of 20 Taiwan Chinese pancreatic adenocarcinoms had K-ras codon 12 mutations. Of 18 mutations, 17 were GGT-GTT mutations. It seems that K-ras GGT-GTT mutation plays an important role in Taiwan Chinese pancreatic carcinogenesis.To our knowledge there were few reports on K-ras mutation pattern of pancreatic adenocarcinoma from Chinese mainland patients. We examined 30 cases of surgically resected formalin-fixed, paraffin-embedded pancreatic adenocarcinoma tissue blocks for the presence of activating point mutations at codon 12 and 13 of the K-ras gene. Pancreatic adenocarcinoma is characterized by its strong desmoplastic and inflammatory reactions. Because of these features, it is very difficult to obtain cancer cell preparations, which are pure enough for molecular analyses.
