Research Of The Effect Of Simiaowan To Knee Joint Oteoarthritis Chondrocyte Apoptosis And Proliferation

Objective: Research the effect of the Simiaowan on chondrocyte apoptosis and multiplication of hind limb of rabbits experimental osteoarthritis.Methods: The experimental osteoarthritis were duplicated by immobilizing with gypsum in extension position in right hind limb of rabbits. Observe the pathological changes of articular cartilage and synovial membrane by HE and Masson colouration under light microscope and scanning electron microscope,then mark it by Mankin. Check chondrocyte apoptosis index(AI) by TUNEL,and Check multiplication index by PCNA.Measure expression rate of bcl-2 and bax by RT-PCR,and measure IL-1βof arthrosis synovia by radioimmunoassay.Rusult: Under light microscope and scanning electron microscope,after treating with Tonifying the Kidey decoction, inflammation of articular cartilage and synovial membrane were lightened, the histological changes of cartilage of model rabbits were relieved, chondrocyte apoptosis index were decrease and multiplication index were steady,expression rate of bcl-2 were raised and that of bax were lowered, IL-1βof arthrosis synovia were decrease.Conclusion:The Simiaowan can restrain chondrocyte apoptosis of knee Oteoarthritis,and relive damnification of articular cartilage. The action may be caused by two approaches,which the first is decreasing grade of cytokdnes in synovia and the second is raising expression rate of bcl-2 and lowering that of bax.
