Effects Of Compound Rhizoma Smilacinus Granule On Apoptosis, Genes Expression Of Bcl-2 And Bax In Non-small Lung Cancer A549 Cell Lines

Objective: To evaluate the effects of Compound Rhizoma Smila-cinus granule (CRSG) on apoptosis , related genes expression of Bcl-2 , Bax in A549 cell lines and its mechanisms upon non-small cell lung cancer by the means of serum pharmacology of Chinese medicine.Methods: The SD rats were randomly differentiated into two groups (n=15/each group), one was administrated with CRSG(twice /d), another with the same dose of distilled water . The serum was obtained two hours after the sixth treatment. Non-small cell lung cancer A549 cell lines were randomly divided into CRSG group , DDP group and normal control group which were cultivated with 10% herbage-containing serum , DDP (3μg/ml) and 10% SD rats serum respectively . The cultivated cells vere collected after 48 hours , then spectrophotometry was used to determine the effective elements — diosgenin of Rhizoma Smilacinus in CRSG-containing serum , electron microscope (TEM) to observe the shape of the cells , flow cytometry assay to detect the cell cycle and the rate of. apoptosis , and RT-PCR technique to determine the expression of Bcl-2 , Bax mRNA.Results:1. The effective elements-diosgenin of Rhizoma Smilacinus in CRSG -containing serum was discovered for the first time .2.
