Objective: To reveal the correlation between somatic mitochondrial DNA oxidative injury and Osteoarthritis. And explore the corelational factors of pathogenesis and pathology course of Osteoarthritis on molecule level.Method: There were seven hip joint cartilage specimens obtained from Osteoarthritis patient and five specimens from normal human. Specimens were divided into Osteoarthritis group and normal group. Cartilage cell nucleolus and somatic mitochondrial DNA was distilled. Consistency and pure degree of the distilled DNA were measured with spectrophotometer. And then the 8-OHdG content of 10μl nDNA and mt DNA were measured with HPLC/EC methods. The Test was used to detect significant changes of the differences in 8-OHdG content of nDNA and mt DNA in each group (a=0.05).Result: In Osteoarthritis group, the content level of 8-OHdG in nDNA and mtDNA of cartilage cells increase significantly compared with normal group (P<0.05 and P<0.01) .There is no significant difference in 8-OHdG content of nDNA(ng/ μgDNA) between the two groups (P>0.05) .
