Objective Single nucleotide polymorphism (SNP) is the majortype of genetic variant in human genome. SNP located within the non-recombinant part of the Y chromosome(NRY) and in the mitochondrial genome possess uniparental inheritance (paternal and maternal, respectively) and do not suffer recombination. It is particularly valuable to establish a high-throughput SNP genotyping method which will be used to supplement STR in forensic casework. The aim of the present study was to establish a rapid and robust assay to simultaneously genotype SNP by the single nucleotide primer extension (minisequencing) with PCR multiplexes. With this method, we obtained the population genetic data of Y-SNP and mt-SNP in Chinese population in Sichuan and the analysis of SNP is a promising application in forensic casework.
