Objective: construct plasmid pcDNA3.1(+)-NKX3.1 with homeobox gene NKX3.1; to transfect DU-145 and explore the effects of NKX3.1 on DU-145; and ascertain the pathways of this effects.Methods: â‘ amplify NKX3.1 sequences from LNCaP cell by PCR. Then construct PcDNA3.1 (+)-NKX3.1 with molecular clonal technique and identify the sequences.â‘¡after plasmids transfected via liposome, observe the effects of NKX3.1 on DU-145 cell line growth, cycle and apoptosis by FCM, MTT and AO EB. Detect the level of IAPl with RT-PCR, compare the differences of DU-145/NKX3.1 and DU-145.Results: â‘ PcDNA3.1(+)-NKX3.1 are constructed and the sequence is correct.â‘¡FCM: G1 stage fraction increased and S stage decreased evidently of DU-145/NKX3.1 (P<0.05). MTT: cell growth of DU-145/NKX3.1 are suppressed.
