Part One: Primary culture and indentification of embryonic cerebralcortical neurons of a rat and establishment of hypoxic injured cell-modelin vitroObjectives: To culture and identify embryonic cerebral cortical neurons of a rat and set up hypoxic injured cell-model of cerebral cortical neurons of a rat embryo in vitro.Methods: (1)Embryonic cerebral cortical neurons of a rat were cultured in vitro by combined method of typspin digestion and DNase I based on the neurobasal medium. (2)The cultured embryonic cerebral cortical neurons were identified with immunofluorescin histochemistical method. Tubulinβmonoclonal antibody and GFAP polyclonal antibody are specific antibodies for neurons and astrocytes respectively. (3) Hypoxic injured neuron cell-model was established in an anaerobic incubator with an anaerobic gas mixed with 85% nitrogen and 5% carbon dioxide and 10% hydrogen.
