| Apoptosis is a special kind of the death style. It is different from the cellular necrosis and the cellular active "suicide" which is regulated by the genes. Myocardial ischemia will occur when the coronary insufficiency cann't meet the needs of the myocardial energy. As soon as the ischemia occurred, myocardial tissues will not only produce the hypoxia and metabolic disturbance, but also cause the accumulation of the toxic metabolic products. It can cause the ischemic injuries and even the cell death if it continue to aggravate. Early reperfusion can raise the number of the death cells , which is in spite of important to the tissues' survival, but the inflammatory reaction and the accumulation of the neutrophils and the produce of the active oxygen can aggravate the cellular injuries. For a long time, the cell death has been regarded to be the sole style of this myocardial cellular death. But more and more studies showed, the myocardial cellular apoptosis participated in the pathological course of the myocardial ischemia-reperfusion injury. The cellular apoptosis is a new problem to the study of the ischemic heart disease. The inhibiting apoptotic gene Bcl-2 and the promoting apoptotic genes Bax, Fas, P53, C-myc can commonly regulate this process. We use the reperfusion model of the isolated cultured myocardial cells in rat to study the apoptosis of the myocardial cells and the express of the Bcl-2, Bax, Fas proteins, We confirmed that the isolated cultured myocardial cells has the apoptosis of the myocardial cells. We get the heart from the SD's newborn mouse and culture the primary myocardial cells according to the method of Mustafa, then generate, 2-3 generation of the experimental myocardial cells to be used, which was filled with nitrogen to the culturing bottle to be caused hypoxia. Then the myocardial cells were incubated under the oxygen to be caused oxygen supply again. We used the mouse-anti-mouse Bcl-2 monoclonal antibody and rabbit-anti-mouse Bax, Fas multiclonal antibody and immunohistochemical SABC method, TUNEL method and FCM method to detect the apoptotic cells and using the transmission electron microscope... |
