Tissue engineering is the product of co-development and combination of modern cell biology, biomaterial science, and engineering. The aim of tissue engineering is to investigate and restore tissue and organ substitutes. The establishment of tissue engineering technique means that human beings will be out of the era of tissue and organ transplantation, and step into the new era to restore tissue and organ. In this study, bone defect repair and bone formation were conducted employing the method of tissue engineering.1. Bone formation potentiality of cultured marrow stem cells in vitro.Method: Rabbit MSCs were cultured in vitro, cell proliferation and ALP activity were observed.Bone formation potentiality of MSCs in mineralized condition was investigated by von-Kossa staining and scanning electronic microscope. Results: 7-8% bone marrow cells of the third passage were alkaline phosphate positive. Cells occupied the whole bottom of cell culture bottle in about 5-6 days, and began to grow in a manner of bilayers after 10 days, at about 14-15th day small white nodules could be seen on the bottom of bottle. When cultured up to 30 days, von-Kossa staining showed there was calcium deposition in the nodules. Observed by scanning electronic microscope, collagen fibers formed, there was mineral deposition on the fibers on 20th day, trabcular like structure formed on 30th day.2.
