Transgene Expression And Antigen Presentation Mechanisms In Intramuscular DNA Vaccination In Combination With Electroporation

In vivo DNA vaccination with electroporation (DNA+EP) can increase immunoginicity of DNA vaccines. But the specific mechanisms remain unknown. In this study, we investigated long term antigen expression and antigen presentation after DNA+EP immunization in hope of elucidating the mechanisms.We had found DNA+EP caused relatively severe damages to skeletal muscles of mice where EP was applied. However, damages to skeletal muscles alleviated with distance away from electronodes. In area around electronodes, electric pulses of EP broke perimysium of muscle bundles and endomysium of muslce fibers, thus released free muslce fibers from muslce bundles.
