Objective: First of all we discussed 3 kinds of methods to prepare chronic GERD animal model remaining complete stomach, intending to provide ideal animal models for studying the mechanism and management of GERD and its complication. Based on the prepared animal models we further studied the expression of heme oxygenase-1(HO-1),NOS2 in the tissue of esophagi and lungs, then by intervention with inhibitor and inductor of HO-1, to explore the effect of HO-1 in the development of GERD.Method: 123 adult male SD rats were chosen to perform experiment. The animal body weight was 230±30g before operation. The rats were divided into 4 groups randomly: sham operated group(SO), gastro-esophageal reflux mainly group(GER),duodeno-esophageal reflux mainly group(DER)and duodeno-gastro-esophageal reflux group(DGER). 20 rats as SO group were anesthetized, the abdomen were cut routinely, inferior segment of esophagus and duodenum were liberated, and then abdomen were closed after 15 minutes. GER model group rats were prepared model by adopting partial pyloric ligation plus cardiomyotomy(GER1) in 9 rats firstly, because of the high postoperative death rate(8/9)then reformed to adopt esophageal fundus anastomosis(GER2) in 20 rats. Esophago-gastro-jejunal side-to-side anastomosis was used to prepare 3 rats (DER1) as DER model group firstly, but because all of 3 rats died after operation so we altered to adopt duodeno-esophageal anastomosis(bile duct hiascent distally) in 21 rats(DER2). Esophago-gastro-duodenal anastomosis(EGDA)was adopted to prepare DGER model in 47 rats. Postoperative survival animals were feeded with granula fodder, weighed per week, observed nutritional state every day.The animals of each group in batch (3~5 per group every time) were killed by femoral vein bloodletting at 1 , 2, 3 and 4 months after surgery.
