The population of the world is ageing. This increase in the elderly population will inevitably mean a rise in the prevalence of age-related cognitive decline such as Alzheimer's disease(AD). Sex-specific incidence rates for AD are higher in aged women than men. Many fundamental researches and some clinical investigations have reported therapeutic and preventive effects of estrogens on AD. Recent evidences are presented for the wider-ranging actions of estrogen in the brain at the cellular metabolism and neurotransmitter levels. Among these the protective mechanisms of center cholinergic nervous system are clear. With the development of molecular genetics, the genetics is involved and found that ApoE is more related to AD. But the reports about the relationship of estrogen and ApoE is seldom and inconsistent. In this research we study the relationship of estrogen and ApoE and their mechanisms in AD using immunohistochemistry method and insitu hybridization method .We also observe the effect of estrogen on praxiology. Hope to explore the mechanism of AD and provide precise theory basis for the treatment of it.1.The establishment of the ovariectomized AD mice model and the effect of estrogen on praxiologic changesIn order to study the mechanism of AD and provide the aim of pharmaceutical therapy, many animal models have been established though each model can only mimic one part of the cerebral pathological and behavioral changes of AD patients.
