In the study, three basic elements of bone tissue engineering are studied, including: cell scaffold, seed cell and cytokine, to explore the synthesis of calcium phosphate nano-particle by mechanochemical method, and the construction of bone scaffold with calcium phosphate bioglass nanocomposite by organic pickling process; to explore the isolation, culture, properties identification and the induction towards osteoblast of placenta-derived mesenchyme stem cell, and study the feasibility of placenta-derived osteoblast used for bone tissue engineering seed cell; to conduct BMP-7 gene transfection of placenta-derived osteoblast with adenovirus as carrier, and develop artificial bone with calcium phosphate nanocomposite by transgenic placenta-derived osteoblast. The results show that constructed bone scaffold with calcium phosphate bioglass nanocomposite has excellent physical properties, histocompatibility, bone induction activity; The placenta-derived osteoblast has typical osteoblast properties, and can be used for seed cell of bone tissue engineering; The hBMP-7 gene transfection is successfully conducted by adenovirus as carrier, and the transgenic osteoblast can well express objective gene, and the bone formation activity in vivo is significantly enhanced; The conducted nano-artificial bone by transformed Ad-hBMP-7 gene tissue engineering has no acute toxicity and hemolytic reaction, and large segment of bone defect at weight-bearing site is repaired successfully in animal test. It is verified that transgenic nano-artificial bone has superior bone replacement function.
