Objective To test the expressions of NgR mRNA suppressing by the vector based siRNA199 cognate to NgR.Methods To construct a recombination plasmid of RNA interference, evaluated the vector by enzyme cutting and gene sequencing test; we introducted recombination plasmid into dissociated postmitotic cultures prepared for cells from the rats' hippocampus and forebrain, studied the vector transfection by observing the marker cGFP gene expressing, evaluated NgR gene suppressing by using real-time PCR;Then we created a model of SCI of rats, followed microinjection the vector based siRNA199 in hindlimb motor area of cerebral cortex. Results: We created a recombination plasmid vector targeting NgR-specific siRNA199 successfully.The vector molecular weight and gene sequence is correct by using enzyme cutting and gene sequencing test. Vector based siRNA199 suppressing NgR gene expression successfully. Conclusions: Vector based RNAi targeting NgR-specific siRNA199 can suppress NgR mRNA expression in cultured rats' cortical and hippocampal cells, can promote neurological functional recovery and axon regeneration of rats after SCI, which indicates that vect base RNAi may be a potential therapeutic method of SCI.
