Objective : According to some criteria of International Organization for Standardization and our country ,we gived a study to the Nature Hydroxyaptite/Chitosan (NHC)material in vivo and in vitro .The purpose of our study was to evaluate the biocompatibility and biological security,to study the mechanism of osetogenesis,to analyze the ability to repair bone defect as the scaffold of bone tissue engineering,to supply the database and reference data for further applying in clinic. Materials and methods: This study had two separate parts in the biological experiment .In the first part our study included acute general toxicity and heat source test and hemolysis test and mutagenicity test in Kunming mice and NewZealand rabbits.The cellular biocompatibility was evaluated through osteoblast from primary culture and human bone marrow derived mesenchymal stem cells.In the second part we used five types of materials: NHC of two sizes (50μm and 500nm) , chitosan ,pure Hydroxyapitite and massive NHC ( 10x2mm).They were implanted into subcurtaneous of the back of 12 healthy adault NewZealand rabbits.The specimens were harvested after operation 1,4,12 and 24 weeks respectively and examined by clinic gross observation and CT and histology and analyzed by computer-assisted image analysis system.The massive material were also put into the parietal bone defect of 22 NewZealand rabbits at random.The rabbits were sacrificed at l,4,12,20,24and 40 weeks postoperation .The specimens were examinated by clinic gross observation and CT and three dimensions reconstruction and histology and SEM.
