Objective Part one: To construct eukaryotic expression vector of humanconnective tissue growth factor (CTGF) gene and obtain the expression results from mammalia Cos7 cell which could promote the proliferation of human umbilical vein endothelial cell(HUVECs). Construction of eukaryotic expression vector successfully laid the foundation for further study on the role CTGF gene playing in angiogenesis and tumor. Part two: (1) To study the effects of TGF β 1, endogenous CTGF and exogenous CTGF on the proliferation, apoptosis and cell cycle of Pancreatic cancer Panc-1. (2) To investigate the expression of CTGF in Panc-1 and the effect of TGF β 1 on the expression of CTGF. (3) To study the effect of anti-CTGF antibody on function TGF β 1 in Panc-1. (4) To investigate the mechanism of promoting Panc-1 apoptosis. According to the results of the above experiments, the relationship of expression of CTGF and proliferation/apoptosis of Pancreatic cancer were investigated. Part three: (1) To observe the effect of CTGF on the proliferation of HUVECs; (2) To investigate the effect of CTGF on extracellular signal-regulated kinase (ERK) and/or phosphatidylinositol-3-kinase(PI-3K) signaling pathways and the effect of LY294002 (a specific PI3K inhibitor) and U0126 (a specific inhibitor of ERK phosphorylation) on function of CTGF in HUVECs.Methods Part one: According to human CTGFcDNA sequence, We designed a pair of specific primers which contained digestion site of XbaI and HindIII on the 5' end...
