Nowadays heart transplantation has become an important therapeutic method of late stage heart failure in clinical practice. However the side effects of non-specific immune suppression remain a big problem. Induction of transplantation tolerance seems to be an ideal route to avoid this problem.The cells-followed-by CP system has been intensively studied in murine and rat models, and it appears to be potentially applicable to human transplantation. However, this tolerance-inducing method cannot overcome fully allogenic(MHC plus mHC) antigen barriers. In this study we used mutiple infusion of donor spleen cells combined with cyclophosphamide(CP) to see whether heart allografts tolerance in rats could be achieved across fully allogenic antigen barriers, meanwhile, we studied the tolerance mechanisms. In order to explore the relationship between microchimerism and transplant tolerance, adult rats parabiosis model was made before heart transplantation to see wheater heart allografts tolerance could be achieved, meanwhile we studied the tolerance mechanisms on cellar and molecular level.
