Both uncontrolling proliferation and invasion ,metastasis are the main traits of malignant tumors; over one hundred tumor-related genes (oncogenes) are involved in growth and spread of neoplasms, and some of these gene expression products are located on cell membrane. Monoclonal antibodies against the tumor-related membrane proteins play an important role in tumor diagnosis, treatment and novel tumor-associated gene discovery.Lou/c rats were immuned with 1.5×10~7 cell of mice dendritic cell sarcoma (DCS) cell line intraperitoneally. Immune spleen cells (2×10~8) were fused with rat myeloma IR983F (1×10~8) by adding 40% PEG1000 solution dropwise into the cell mix. A monoclonal antibody having no cross immune reaction with mice normal tissues was produced and named 983 D4. The cell membrane antigen recognized by 983 D4 is also expressed in some other mice cancers such as gastric cancer MFC, lung cancer LA795, glioma B22 and its molecular weight is about 38kD, 28kD. Effects of 983 D4 on biological behaviors of DCS cell were investigated. It was indicated that 983 D4 can inhibit cell in vitro growth and colony-forming efficiency, but had no significant effects on cell motility, lysis, adhesion, invasiveness and metastatic potential. DCS cell cDNA expression library in λgt11 was contracted and the library was screened with 983 D4 in order to further investigate the antigen feature.1.13mg of total RNA of DCS cells was isolated by a single extraction with an acid guanidinium thiocyanate-phenol-chloroform mixture.
