This study was designed to determine the incidence and characteristics of the mutant of the tumor suppressor gene p53 in Chinese lung cancer patients (NSCLC & SCLC) and to explort the possible association between the mutant of the p53 gene and the c 1 inicopatholgic feature, especially the postopertive prognosis. Data thusobtained might be served as a guid to predict thelong- term outcome to install postoperativeadjuvant therapy. Monoclonal antibody to human p53protein (pAbl801 & DO-1) and immunohistochemicalmethods were used to detect p53 proteinaccumulation in formalin-fixed, paraffin-embeddedas well as fresh-frozen tissue specimens of lungcancer (202 cases of NSCLC and 32 cases of SCLC). 121 out of 202 NSCLC (59.9%)were p53 positive, including 63 out of 98 (64.3%) squamous carcinoma,41 out of 75 (54.7%) adenocarcinoma, 16 out of 26(61.5%)adenosquamous carcinoma and 1 out of 3 large cell carcinoma. Nuclear p53 protein was also observed in 81. 3% of 32 SCLC. ABC is a simple, effective, accurate and reliable method of screening the p53 protein overexpression of lung cancer.
