Xenotransplantation is an effective pathway to improve the shortage of graft, however the intense xenograft rejection greatly hindered its application in clinic. Accommodation being a state between rejection and tolerance had obtained widespread attention, and been considered to take as a pathway to overcome acute vascular rejection (AVR) in xenotransplantation. This research is based on above understanding. In the study, the PAEC (Porcine Arteriae Endothelial Cell) was cultured and expanded in vitro, reduced to develop the state of accommodation with different ways. Then we cloned the differentially- expressed genes in accommodation PAEC by using PCR-based and improved subtractive hybridization, and analysis the biological response that maybe proceed in the accommodation PAEC to find the clue of studing the accommodation developed mechanism.
