OBJECTIVESTo elucidate the regulatory effects of five NF-κB sites (-898 to -888bp, -641 to -632bp, -610 to -601bp, -498 to -488bp, -208 to -198bp) in rat TNF-α promoter region on Angiotensin II(AngII) induced TNF- α gene transcription in cardiac myocytes of neonatal rats.MATERIALS AND METHODSDifferent upstream sequences of TNF- α gene(-1167 ~ +63bp, -730 ~ +63bp, -632 ~ +63bp, -548 ~ +63bp, -421 ~ +63bp) were fused to the luciferase reporter gene, promoterless pGL3-basic vector, and transiently cotransfected with the pRL-TK vector into neonatal rat myocytes.
