CiSPLatin can still be considered the most versatile platinum-containing antineoplastic drug. Besides nephrotoxicity, ototoxicity is one major dose-limiting side-effect of the drug, and it can lead to irreversible sensorineural hearing loss. The most severe damage occurs in the third row of outer hair cells at the basal turn of the organ of Corti. However, details of the molecular mechanism have not yet been clarified. MAPK family includes three major members: JNK/cjun, p38 and ERK, one of their major roles is to act as a mediator of apoptosis, JNK/cjun and p38MAPK are probably crucial in transmitting the signals from the extracellular enviroment to the nuclear transcriptional machinery when apoptosis is occurring, while activated ERK1/2 is thought to have several actions, such as supporting the survival of cells after noxious influences. OBJECTIVE1 This study was designed to establish an experimental cddp ototoxity model in guinea pig, which could exhibit inner ear cell morphology, ultramicro structure and function and to study the mocular mechanisms.2 To investigate whether apoptosis is one of the mechanism for cell death in spiral ganglion cells(SGC) and organ of Corti (OC) cells of guinea pigs'cochlea induced by ciSPLatin.3 To investigate the expression of JNK/cjun protein and cjun mRNA, and pp38MAPK and pERK1/2, regulators of apoptosis, in guinea pig cochlea injuried by ciSPLatin.MATERIAL AND METHOD1.
