| Glaucoma is the major cause of blindness in the world as well as in China. Exploring the pathogenesis, making early diagnosis and treatment for glaucoma are the important fields in glaucoma research. ELAM-1 is the only molecular marker found in glaucoma patients. The study of ELAM-1 in glaucoma may be helpful for further understanding the pathogenesis.Until now, ELAM-1 is mainly studied in vascular diseases. Since there exists a similarity between vessels and trabecular meshwork, the techniques applied in the vascular research were adopted to evaluate the expression and function of ELAM-1 in trabecular cells, and the changes of the signal transduction in the process of the production and function of ELAM-1, to elucidate the role of ELAM-1 in the pathogenesis of glaucoma. Our study is consisted of three parts:Part I The study of expression of ELAM-1 on the aqueous outflow pathway. Objective Oxidative stress and high blood pressure are the main risk factors of vascular diseases. Oxidative stress and high intraocular pressure (IOP) are also main risk factors in glaucoma. The expression of ELAM-1 in vascular diseases is related to oxidative stress and high blood pressure. Therefore, we evaluate the effect of oxidative stress and high IOP on the expression of ELAM-1 in trabecular cells, and evaluate if the expression of ELAM-1 is regulated by IL—1a, in order to find the mechanism of the expression of ELAM-1 in the glaucomatous trabecular cells. Material and Methods After 16 hours of serum starvation, primary cultured porcine trabecular meshwok cells (PTC) were divided into three groups: control group, H2O2 group and IL-1ra group. IL-1ra group is again divided into subgroups depending on the concentration of IL-lra added. PTCs in the control group were cultured with serum free media. PTCs in H2O2 group were treated by 1mM H2O2.
