γδ T cells have dual-feature of innate and acquired immunity. However, the exact role of γδ T cells in immune system is not clear yet. Clarification of the mechanism of antigen recognition by γδ T cells is not only essential for better understanding biological function of γδ T cells, but also is important to clarify the regulatory mechanism of innate immunity to acquired immunity. Consequently, it will promote the further understanding of the essence of immune response and also form novel strategy for immune therapy.The present study focus on the antigen recognition mechanism of γδ TCR to HSPs or TCR Vδ2 to HBV infected tissue/cells. Then synthesized CDR3δ peptide was employed to screen for and identify peptides recognized by γδ T cells. 1. Study on the antigen recognition mechanism of human γδ T cells to membrane HSPsHeat shock proteins (HSPs) play key role in fundamental immunological phenomena such as activation of APCs and chaperoning of peptides during antigen presentation or as possible antigens to initiate and drive immune responses. Therefore, in present study we aimed to make clear the exact role of HSPs when interacted with γδ T cells, whether as antigen presentation molecule or as antigen directly? Furthermore, what is the biological significance of thus interaction, especially in tumor immunosurveillance? We used EBV transformed B cells (LCL cells) as model to study effect of stressful events caused by cell transformation on HSPs expression on cell membrane. In order to investigate whether HSPs were involved in the response of human γδT cells to transformed cells, the cytotoxicities of γδ T cells to transformed cells with or without an HSP60/70 gene knockdown were evaluated. Then, proliferation assays were performed to evaluate whether HSPs could stimulate γδT cell in vitro directly.
