Part I Construction of Human Glioma Tissue MicroarrayObjective: To construct human glioma tissue microarray, for the purpose of providingexperimental platform for glioma associated researches.Methods: Two 118-spot tissue microarray were constructed by domestic tissue arraysample spotting instrument half-manually with some modification.Results: Microarray were designed according to the requirement of specimen.Recipient paraffin blocks were prefabricated firstly, then with localization of donatorparaffin blocks, sampling, slicing continuously, supporting plates and parching platessequentially. Finally two 36*26mm sized paraffin blocks were prepared, each had 118spots and each spot was 1.5mm in diameter, microarray were cut into 4μm per layer. Pointlattices distributed evenly under macroscopic observation, while 4 point lattices displaced,displacement ratio was 0.03%. HE slices showed sharp margin surrounding samplingtissue without obvious tissue crushing and deformation.Conclusions: 1. Two tissue microarrays were constructed, which were composed ofhuman gliomas, other intracranial tumors, xenograft tumors, genetic engineered mousebrain tumors, normal brain tissues and other human normal tissues. These arrays can offerhigh quality experimental platforms for glioma associated researches with high efficiencyand convenience. 2. Preparation of high quality tissue array relied on standardizedspecimen paraffin block libraries. 3. reliability of tissue array experiments depends onaccurate pathological diagnosis of donator blocks and exact corresponding positioning. 4.Beeswax can be added to donator blocks to a final concentration of 2.5%, in order to reacha well balance between rigidity and flexibility.
