The essential biological materials-serum albumin and deoxyribo- nucleic acid (DNA) have been playing many vital roles for all kinds of biological phenomena. Exploring the interaction mechanisms on these bio-macromolecules with small molecules or ions, especially for those targeting-drug molecules, at the molecular level is of current interest in many research area such as biology, clinical medicine, medicinal chemistry, chemistry and so on. As a part of"Thermodynamics and Mechanism of Interaction between Bio-macromolecules and Antibiotics", Which was supported by National Natural Sciences Foundation (No.: 20173050), the present work consists of the following several parts.1. In the first part, the binding characteristics and mechanism of quinolone antibiotics to the bovine serum albumin (BSA) have been investigated by using fluorescence spectroscopy and absorbance spectra. Based on the site binding model and fluorescence quenching, a modified formula that can be used to calculate the binding parameters of small molecule ligand binding to bio-macromolecule has been proposed. Further more, the equilibrium constants and number of binding sites for these quinolone drugs in this research interacting with BSA are measured by fluorescence method, the transfer efficiency of energy and distance between these drugs and BSA are obtained in view of the theory of F?rster energy transfer, the effects of each drug on the conformation of BSA are investigated by synchronous fluorescence spectra and allosteric domain model of drug-BSA complexes are postulated.2. In this part, the technology of three-dimensional fluorescence spectrometry (TDFS) is developed and has been used to study the conformation change of BSA after the binding interaction of these quinolone drugs with BSA.
