| Hypoxia is the one of the main causes of radioresistance in malignant tumors. It is hypothesized that methods which can reduce the extend of hypoxia in tumors would improve the effects of radiotherapy. Sanazole(SAN) is a radial chemical modifier of nitrotrizoles. Former studies showed that it possesses strong radiosensitizative effects on some tumors. It is made by our groups using a new method and is now being studied in phase I trial.OBJECTIVE To study the radiosensitive effects and mechanism underlying radiosensitization of sanazole, as well as its safty.METHODS Part A: Observation of the growth rate of cultured breast cancer cell (MCF7) and cervical carcinoma cell (HeLa) treated by SAN with 60Co γ radiation in hypoxia. Part B: Using flow cytometry, the cell cycle distributions and apoptosis of cultured HeLa cells were checked in 24h after treated by SAN combined with y -radiation. The proteomic analysis of the HeLa cells in 24hr after being treated by SAN with γ -radiaion were performed by two-dimensional gel electrophoresis(2-DE) and annlazed by imagemaster 2Dplatinum software.
