Objective:1. To investigate gene expression profiles during various stages of embryonic pancreatic development of rats, and determine the key genes relevant to insulin synthesis and exocytosis.2. To study the difference of insulin secretion and its mechanisms in normal rats and the rats with intrauterine growth retardation.Methods:1. Pancreata of SD rat at embryonic day 12.5(E12.5), E15.5, E18.5 and newborn (N), 21 days after birth (P21), adult (A) rats were dissected respectively.2. Genechips from Affymetrix company were applied to explore gene expression profiles. The data were then analyzed by bioinformatical method. Some genes related to insulin synthesis and secretion were verified by RT-PCR and realtime PCR.3. RT-PCR, Western blot, immunohistochemistry, immunofluore-scence were used to study the expression of genes and proteins relavant to insulin release.4. Intraperitoneal glucose tolerance tests (IPGTT) and ELISA were done to detect the function of pancreatic islet.
