To recover the structure and function of damaged endothelium as much as possible and to block or to delay the evolvement of the endothelium damage are the urgent problems in the treatment of these diseases.As one of the important treatment methods of ischemic disease, angioplasty (or revascularization) is becoming more popular for it's less injury, definite effect and quick recovery. By the dilation, it can press or remove the proliferating tissue to open the vessel and recover the blood flow. As a intraluminal operation, revascularization will unavoidably damage the endothelium and maybe cause the restenosis (RS) which is the major complication of angioplasty. Even in the era of stenting, the occurrence rate of restenosis is still as high as 12%-23%. The mechanism of RS is still unkown, but there were studies showed that the balloon dilation damage of the vessel endothelial barrier, exposure of the vessel stroma, activation, phenotype transformation and proliferation of smooth muscle cells (SMCs), unbalance of extracellular matrix (ECM), persistent thickening of vessel intima are related to the RS. At present the drug eluting stent and intravascular radiation have good effect on the RS, but these methods will inhibit the recovery of endothelium and have the risk of late thrombosis. How to protect the endothelium, reduce the damage of endothelium, speed up the reestablished endothelial integrity of target vessels to reduce the rate of RS? It is a very important topic in angioplasty and the study of stem cell give us hope to initiate a brand new way to treat the ischemic disease.Stem cells are defined as cells that have self-renewing capacities and can differentiate into multiple cell lineages and can differentiate into different kinds of function cells to form many types of tissue and organs. According to the different order in the ontogenetic process, the stem cell can be divided into embroynic stem cells(ESCs) and adult stem cells(ASCs).
