Mesenchymal stem cells(MSCs) are multipotential stem cells derived from mesoderm of early phase embryo. Recent studies showed that MSCs possessed immunomodulatory potential. So it may be used to induce immuno-tolerance in organ transplantation. In the study, we isolated MSCs from rat bone marrow and established a model of heterotopic rat heart transplantation to research the effect of tolerance induction with MSCs on rat heart transplantation and to explore the underlying mechanism.In part I of the research, we successfully isolated MSCs from rat bone marrow by ingredient centrifugalization. The cells were purified by adhering speciality. In part II, we choosed Sprague-Dawley(SD)rat as recepient and Wistar rat as donor, to establish a heterotopic model of rat heart transpalntation with more than 95 percent of the transplantation successful. In part III, we explored the effect of MSCs on mixed lymphocyte reaction(MLR) in vitro by 3H-TDR incorporating method. The results showed that MSCs would not elicit a proliferative response of rat lymphocyte, but had significant suppressive effect on alloreactivity of rat lymphocyte. In part IV, we investigated the influence of MSCs on allograft of rat heart transplantation. In this part, all recepients were divided into four groups. In group I, recepients received 1ml PBS via tail vein after transplantation. Recepients in group II received 1*106/1ml MSCs via tail vein.
