| Objective To investigate the optimal time for mesenchymal stem cells transplantation on myocardial infarction.Method Adult inbred strain Wistar rats were randomly divided into sham group (n=6) and model groups (n=79). Rats in the latter group were established model of myocardial infarction by ligation of the anterior descending branch of the left coronary artery. Rats in the model group were compared with rats in the sham group on ECG, HE staining tissue sections and heart function to evaluate the model. Then rats established the model of myocardial infarction successfully were divided randomly into bone marrow collecting group (n=6), DMEM group (n=24) and MSC group (n=24). After separated, cultured and proliferated mesenchymal stem cells collected from the femurs, tibias and humerus of rats in the bone marrow collecting group, 1×10~6 mesenchymal stem cells (0.15ml) were transplanted to the rats in MSC group one hour (MSC-1 group, n=8), two weeks (MSC-2 group, n=8) or four weeks (MSC-3 group, n=8) after myocardial infarction respectively. Culture medium (0.15ml) were transplanted to the rats in DMEM group one hour (DMEM-1 group, n=8), two weeks (DMEM-2 group, n=8) or four weeks (DMEM-3 group, n=8) after myocardial infarction as the same way. Four weeks after cell transplantation, left ventricular function was tested and specimens of ischemic myocardium from every group were got respectively for assaying the survival and differentiation of transplanted cells as well as angiogenesis.Result Rats established model of myocardial infarction have... |
