| Spirodela polyrrhiza (L.) Schleid, which distributes widely in china, is a kind of traditional Chinese herbal medicine with a long history of widely used to treat urticaria, acute nephritis, et al. Some reports indicate that Spirodela polyrrhiza (L.) Schleid has the cancer preventive potential. But, there is no any report on the research and utilization of anti-cancer compounds in Spirodela polyrrhiza (L.) Schleid. The object of this paper is to determine the anti-cancer compounds, optimize the extraction and purification technology of these compounds from Spirodela polyrrhiza (L.) Schleid, estimate the quality stability of extract, and study the cancer preventive effect and the protective effect on hydrogen peroxide induced cells damage of the compounds. The major contents and results of the research are as follows.The 75% ethanol extract of Spirodela polyrrhiza (L.) Schleid was partitioned successively with petroleum ether (PE), ethyl acetate (EtoAc), n-butanol (n-BuOH) and the residues of aqueous. Then, the combination of EtoAc and n-BuOH was separated to 8 parts by AB-8 macroporous adsorption resin. The total flavonoids content and the inhibitory effect on human hepatocellular carcinoma cell line SMMC-7721of each part were determined. The results showed that the part which had the higher flavonoids content, inhibited SMMC-7721 growth more potently. The results suggested that the total flavonoids in Spirodela polyrrhiza (L.) Schleid was the major bioactivity part of anti-cancer.The extraction technology for total flavonoids in Spirodela polyrrhiza(L.) Schleid was systematically studied with single factor experiment and orthoganol design. The conventional optimum technological conditions were attained as followed: extracted at 60 ℃ with 60% ethanol, the ratio of materiel to solvent was 1:10, and extracted for 2 times, 2h for each time.The flocculation with chitosan method was studied in this paper, and the results of single factor experiments showed that, when the temperature of flocculation was 60℃, the chitosan content was 6% of the volume of the extraction solution, the pH was about 5.5 and the viscosity of chitosan was 30 Pa.s, the effect of flocculation was better.The purification of total flavonoid by using the method of macroporous adsorption resin was experimented.
